    Formation des PE2 à l'élaboration et la conduite d'un projet patrimoine et citoyenneté

    Comment former de jeunes collègues PE2 à la pratique d'une pédagogie de projet centré sur la citoyenneté et le patrimoine ? Telle est la problématique de formation développée ici.Ce projet, mené sur le site IUFM de Nîmes en 2001-2002, a associé 8 stagiaires PE2 qui ont travaillé en équipe avec des professeurs de l'IUFM, un IMF et des intervenants extérieurs ; le projet pédagogique a été mené sous la forme d'interventions « filées» dans une classe de CM1 de l'école Paul Marcellin de la ZUP de Nîmes. Pour aider les élèves à construire leur identité à partir de la question « Qu'est-ce qu'être Nîmois?», les stagiaires les ont amenés à découvrir leur ville en partant sur les traces d'un Nîmois célèbre, Alphonse Daudet.How to train young student teachers in the conduct of a pedagogical project focussed on civics and cultural/historical heritage? Such are the issues developed in this article.This teacher training project, conducted on the Nîmes campus of the Montpellier teacher Training College (IUFM) over the 2001-2002 school year, brought together 8 trainees who teamed up with some IUFM professors, an IMF, as well as outside participants. The conduct of this project took the form of regularly scheduled pedagogical participation by the trainees in the instruction of a clans at a school located in the Nîmes ZUP (urban renewal project). The teaching project consisted of working on the theme of identity on the basis of the question: "What does it mean to be Nîmois?" In order to answer this question, the trainees made the pupils discover the city of Nîmes by following the footsteps of a famous Nîmois, Alphonse Daudet

    Stress Clamp Experiments on Multicellular Tumor Spheroids

    The precise role of the microenvironment on tumor growth is poorly understood. Whereas the tumor is in constant competition with the surrounding tissue, little is known about the mechanics of this interaction. Using a novel experimental procedure, we study quantitatively the effect of an applied mechanical stress on the long-term growth of a spheroid cell aggregate. We observe that a stress of 10 kPa is sufficient to drastically reduce growth by inhibition of cell proliferation mainly in the core of the spheroid. We compare the results to a simple numerical model developed to describe the role of mechanics in cancer progression.Comment: 5 pages, 4 figure

    Bioinformatic analysis of ESTs collected by Sanger and pyrosequencing methods for a keystone forest tree species: oak

    <p>Abstract</p> <p>Background</p> <p>The Fagaceae family comprises about 1,000 woody species worldwide. About half belong to the <it>Quercus </it>family. These oaks are often a source of raw material for biomass wood and fiber. Pedunculate and sessile oaks, are among the most important deciduous forest tree species in Europe. Despite their ecological and economical importance, very few genomic resources have yet been generated for these species. Here, we describe the development of an EST catalogue that will support ecosystem genomics studies, where geneticists, ecophysiologists, molecular biologists and ecologists join their efforts for understanding, monitoring and predicting functional genetic diversity.</p> <p>Results</p> <p>We generated 145,827 sequence reads from 20 cDNA libraries using the Sanger method. Unexploitable chromatograms and quality checking lead us to eliminate 19,941 sequences. Finally a total of 125,925 ESTs were retained from 111,361 cDNA clones. Pyrosequencing was also conducted for 14 libraries, generating 1,948,579 reads, from which 370,566 sequences (19.0%) were eliminated, resulting in 1,578,192 sequences. Following clustering and assembly using TGICL pipeline, 1,704,117 EST sequences collapsed into 69,154 tentative contigs and 153,517 singletons, providing 222,671 non-redundant sequences (including alternative transcripts). We also assembled the sequences using MIRA and PartiGene software and compared the three unigene sets. Gene ontology annotation was then assigned to 29,303 unigene elements. Blast search against the SWISS-PROT database revealed putative homologs for 32,810 (14.7%) unigene elements, but more extensive search with Pfam, Refseq_protein, Refseq_RNA and eight gene indices revealed homology for 67.4% of them. The EST catalogue was examined for putative homologs of candidate genes involved in bud phenology, cuticle formation, phenylpropanoids biosynthesis and cell wall formation. Our results suggest a good coverage of genes involved in these traits. Comparative orthologous sequences (COS) with other plant gene models were identified and allow to unravel the oak paleo-history. Simple sequence repeats (SSRs) and single nucleotide polymorphisms (SNPs) were searched, resulting in 52,834 SSRs and 36,411 SNPs.     Comparing HLA Shared Epitopes in French Caucasian Patients with Scleroderma

    Although many studies have analyzed HLA allele frequencies in several ethnic groups in patients with scleroderma (SSc), none has been done in French Caucasian patients and none has evaluated which one of the common amino acid sequences, 67FLEDR71, shared by HLA-DRB susceptibility alleles, or 71TRAELDT77, shared by HLA-DQB1 susceptibility alleles in SSc, was the most important to develop the disease. HLA-DRB and DQB typing was performed for a total of 468 healthy controls and 282 patients with SSc allowing FLEDR and TRAELDT analyses. Results were stratified according to patient’s clinical subtypes and autoantibody status. Moreover, standardized HLA-DRß1 and DRß5 reverse transcriptase Taqman PCR assays were developed to quantify ß1 and ß5 mRNA in 20 subjects with HLA-DRB1*15 and/or DRB1*11 haplotypes. FLEDR motif is highly associated with diffuse SSc (χ2 = 28.4, p<10−6) and with anti-topoisomerase antibody (ATA) production (χ2 = 43.9, p<10−9) whereas TRAELDT association is weaker in both subgroups (χ2 = 7.2, p = 0.027 and χ2 = 14.6, p = 0.0007 respectively). Moreover, FLEDR motif- association among patients with diffuse SSc remains significant only in ATA subgroup. The risk to develop ATA positive SSc is higher with double dose FLEDR than single dose with respectively, adjusted standardised residuals of 5.1 and 2.6. The increase in FLEDR motif is mostly due to the higher frequency of HLA-DRB1*11 and DRB1*15 haplotypes. Furthermore, FLEDR is always carried by the most abundantly expressed ß chain: ß1 in HLA DRB1*11 haplotypes and ß5 in HLA-DRB1*15 haplotypes

    Characterization of a cinnamoyl-CoA reductase 1 (CCR1) mutant in maize: effects on lignification, fibre development, and global gene expression

    Cinnamoyl-CoA reductase (CCR), which catalyses the first committed step of the lignin-specific branch of monolignol biosynthesis, has been extensively characterized in dicot species, but few data are available in monocots. By screening a Mu insertional mutant collection in maize, a mutant in the CCR1 gene was isolated named Zmccr1–. In this mutant, CCR1 gene expression is reduced to 31% of the residual wild-type level. Zmccr1– exhibited enhanced digestibility without compromising plant growth and development. Lignin analysis revealed a slight decrease in lignin content and significant changes in lignin structure. p-Hydroxyphenyl units were strongly decreased and the syringyl/guaiacyl ratio was slightly increased. At the cellular level, alterations in lignin deposition were mainly observed in the walls of the sclerenchymatic fibre cells surrounding the vascular bundles. These cell walls showed little to no staining with phloroglucinol. These histochemical changes were accompanied by an increase in sclerenchyma surface area and an alteration in cell shape. In keeping with this cell type-specific phenotype, transcriptomics performed at an early stage of plant development revealed the down-regulation of genes specifically associated with fibre wall formation. To the present authors’ knowledge, this is the first functional characterization of CCR1 in a grass species

    SPICAV on Venus Express: Three spectrometers to study the global structure and composition of the Venus atmosphere

    Spectroscopy for the investigation of the characteristics of the atmosphere of Venus (SPICAV) is a suite of three spectrometers in the UV and IR range with a total mass of 13.9 kg flying on the Venus Express (VEX) orbiter, dedicated to the study of the atmosphere of Venus from ground level to the outermost hydrogen corona at more than 40,000 km. It is derived from the SPICAM instrument already flying on board Mars Express (MEX) with great success, with the addition of a new IR high-resolution spectrometer, solar occultation IR (SOIR), working in the solar occultation mode. The instrument consists of three spectrometers and a simple data processing unit providing the interface of these channels with the spacecraft. A UV spectrometer (118-320 nm, resolution 1. 5 nm) is identical to the MEX version. It is dedicated to nadir viewing, limb viewing and vertical profiling by stellar and solar occultation. In nadir orientation, SPICAV UV will analyse the albedo spectrum (solar light scattered back from the clouds) to retrieve SO2, and the distribution of the UV-blue absorber (of still unknown origin) on the dayside with implications for cloud structure and atmospheric dynamics. On the nightside, 7 and 6 bands of NO will be studied, as well as emissions produced by electron precipitations. In the stellar occultation mode the UV sensor will measure the vertical profiles of CO2, temperature, SO2, SO, clouds and aerosols. The density/temperature profiles obtained with SPlCAV will constrain and aid in the development of dynamical atmospheric models, from cloud top (similar to 60 km) to 160 km in the atmosphere. This is essential for future missions that would rely on aerocapture and acrobraking. UV observations of the upper atmosphere will allow studies of the ionosphere through the emissions of CO, CO+, and CO2+, and its direct interaction with the solar wind. It will study the H corona, with its two different scale heights, and it will allow a better understanding of escape mechanisms and estimates of their magnitude, crucial for insight into the long-term evolution of the atmosphere. The SPICAV VIS-IR sensor (0.7-1.7 mu m, resolution 0.5-1.2 nm) employs a pioneering technology: an acousto-optical tunable filter (AOTF). On the nightside, it will study the thermal emission peeping through the clouds, complementing the observations of both VIRTIS and Planetary Fourier Spectrometer (PFS) on VEX. In solar occultation mode this channel will study the vertical structure of H2O, CO2, and aerosols. The SOIR spectrometer is a new solar occultation IR spectrometer in the range lambda=2.2-4.3 mu m, with a spectral resolution lambda/Delta lambda > 15,000, the highest on board VEX. This new concept includes a combination of an echelle grating and an AOTF crystal to sort out one order at a time. The main objective is to measure HDO and H2O in solar occultation, in order to characterize the escape of D atoms from the upper atmosphere and give more insight about the evolution of water on Venus. It will also study isotopes of CO2 and minor species, and provides a sensitive search for new species in the upper atmosphere of Venus. It will attempt to measure also the nightside emission, which would allow a sensitive measurement of HDO in the lower atmosphere, to be compared to the ratio in the upper atmosphere, and possibly discover new minor atmospheric constituents.     The interest of gait markers in the identification of subgroups among fibromyalgia patients

    <p>Abstract</p> <p>Background</p> <p>Fibromyalgia (FM) is a heterogeneous syndrome and its classification into subgroups calls for broad-based discussion. FM subgrouping, which aims to adapt treatment according to different subgroups, relies in part, on psychological and cognitive dysfunctions. Since motor control of gait is closely related to cognitive function, we hypothesized that gait markers could be of interest in the identification of FM patients' subgroups. This controlled study aimed at characterizing gait disorders in FM, and subgrouping FM patients according to gait markers such as stride frequency (SF), stride regularity (SR), and cranio-caudal power (CCP) which measures kinesia.</p> <p>Methods</p> <p>A multicentre, observational open trial enrolled patients with primary FM (44.1 ± 8.1 y), and matched controls (44.1 ± 7.3 y). Outcome measurements and gait analyses were available for 52 pairs. A 3-step statistical analysis was carried out. A preliminary single blind analysis using k-means cluster was performed as an initial validation of gait markers. Then in order to quantify FM patients according to psychometric and gait variables an open descriptive analysis comparing patients and controls were made, and correlations between gait variables and main outcomes were calculated. Finally using cluster analysis, we described subgroups for each gait variable and looked for significant differences in self-reported assessments.</p> <p>Results</p> <p>SF was the most discriminating gait variable (73% of patients and controls). SF, SR, and CCP were different between patients and controls. There was a non-significant association between SF, FIQ and physical components from Short-Form 36 (p = 0.06). SR was correlated to FIQ (p = 0.01) and catastrophizing (p = 0.05) while CCP was correlated to pain (p = 0.01). The SF cluster identified 3 subgroups with a particular one characterized by normal SF, low pain, high activity and hyperkinesia. The SR cluster identified 2 distinct subgroups: the one with a reduced SR was distinguished by high FIQ, poor coping and altered affective status.</p> <p>Conclusion</p> <p>Gait analysis may provide additional information in the identification of subgroups among fibromyalgia patients. Gait analysis provided relevant information about physical and cognitive status, and pain behavior. Further studies are needed to better understand gait analysis implications in FM.</p

La diffusion a été ainsi étudiée dans des matériaux modèles : composition bien contrôlée, état d’équilibre, absence de gradient de potentiel chimique. La composition chimique interfaciale et la structure particulière qui en découle sont les facteurs essentiels : la diffusion est soit accélérée, soit ralentie notamment quand un atome est rejeté de l’interface. La diffusion chimique est moins bien comprise. L’accélération de la diffusion est d’autant plus importante que le |ΔG| de la transformation est plus élevé.
